Specimen Requirements

Amniotic fluid: 20-30 ml of amniotic fluid obtained by amniocentesis and split into two sterile conical centrifuge tubes. Discard the first 2 ml to avoid maternal contamination.
Chorionic Villus Sampling (CVS): 15-20 mg of villi placed into 15 ml sterile centrifuge tube containing 10 ml transport media provided by DMC UL cytogenetic Lab.
Products of Conception: 500 mg of villi, amnion, or chorion are preferred. Fetal parts should be identified. Place tissue into a sterile tube containing 10 ml transport media provided by DMC UL cytogenetic Lab. Do not freeze tube.
 
Peripheral Blood: 5-10 ml of peripheral blood placed into a green-topped sodium heparin tube. Invert tube several times to mix blood with heparin. Never centrifuge or freeze the tube.
Fetal Blood and Newborn Blood: 1-3 ml of blood placed into a green-topped sodium heparin tube. Invert tube several times to mix blood with heparin. Never centrifuge or freeze the tube.
Bone marrow and leukemic blood: 1-3 ml bone marrow aspirate and 10 ml of leukemic blood with ≥10% blasts placed into a green-topped sodium heparin tube. Invert tube several times to mix sample with heparin.  Never centrifuge or freeze the tube.
Tumor Tissue: 250-500 mg fresh tumor placed into 15 ml sterile centrifuge tube containing 10 ml transport media provided by DMC UL cytogenetic Lab.

aCGH+SNP:  3ml whole blood in lavender top (EDTA) tube AND 3ml whole blood in (Sodium Heparin) green top tube. 1-2mls in each tube is acceptable for neonates 6 months and under.  For prenatal and cancer aCGH see test menu for more details.

Urine for Bladder Cancer UroVysion FISH:  Collect a minimum of 40 ml of voided urine in a sterile container provided by the Cytogenetic Lab.  Cap tightly and invert urine cup several times to mix with preservative tablet.  For specimens where a collection container kit is not available; collect the urine in a sterile container and mix with an equal amount of 70% ethanol.  If unable to ship immediately, refrigerate overnight and ship within 24 hours.  The preferred storage and shipping conditions are on ice packs, but specimens may be stored and shipped at room temperature up to 25°C (77°F).
Paraffin-Embedded Tissue for HER2 Amplification by FISH:  Prepare slides from formalin-fixed, paraffin-embedded tissue block.  Cut 4 consecutive sections at 4-6µm using microtome.  4µm is the preferred thickness for the section.  One of these four sections will be for an H&E slide.  A qualified pathologist should identify and circle the area of invasive tumor with a marker on the H&E slide.
